Former Labor leader in the Legislative Council Adam Searle is on the way out, while Shaoquett Moselmane has also informed the party he will not run.
NSW Labor's right faction on Thursday voted on who it will take to the party's conference for endorsement next month ahead of the March state election.
Current upper house MP Courtney Houssos, Dubbo barrister Stephen Lawrence, Canterbury-Bankstown mayor Khal Asfour, Emily Suvaal and Sarah Kaine have been selected by the right faction.
The left faction has already preselected its candidates, with Labor's final ballot to be determined at the NSW party's annual conference next month.
Mr Searle has been in the upper house for more than 11 years and was aligned with former Labor leader Jodi McKay, who was ousted by Chris Minns just over a year ago.
Mr Minns and Mr Searle are understood not to be close, although they are also not at odds with each other.
Another upper house MP, Walt Secord, confirmed last month he would not re-contest his position, leaving another vacancy.
Mr Secord stepped down from his various roles, including as opposition spokesman for police and the arts, when allegations of bullying were levelled at him following the publication of a report into misconduct in NSW parliament.
He apologised at the time, saying he could be too blunt in a fast-paced work environment.
